Studies on Gamete Formation of Paeonia Japonica MIYABE


Abstract
The pollen tube growth, spermatogenesis and the viability of preserved pollen of Paeonia were examined and following results were obtained.
(1) The pollen of Paeonia, cultured on artificial media in incubator maintained at 25¡É for one hour and twenty minutes showed pollen tube germinated.
(2) 5 to 10% of generative nucleus were moored into pollen tube since the pollen cultured for 5 hours.
(3) 70 to 80% of generative nucleus were mooed into pollen tube since the pollen cultured for 8 hours and its size indicated maximum in length.
(4) In generative cell division, prophase appeared since the pollen cultured for 12 to 20 hours and metaphase was found since the pollen cultured for 25 hours and its chromosome number was 5.
(5) Anaphase was found since the pollen cultured for 30 hours and generative nucleus was divided two.
(6) The pollen of Paeonia may have more than 60% of viability even kept under room condition for 17 days and kept in refrigerator at 4¡É for 120 days.
